Compensatory Islet Response to Insulin Resistance Revealed by Quantitative Proteomics

Abstract

Compensatory islet response is a distinct feature of the pre-diabetic insulin resistant state in humans and rodents. However, the proportional contribution of altered mass versus function per unit mass to sustain the adaptive response is not fully understood. To identify alterations in proteins that characterize the adaptive response, we analyzed islets from five-month-old male control, high-fat diet fed (HFD) or obese ob/ob (OB) mice by liquid chromatography coupled with mass spectrometry (LC-MS). Using global label-free quantification with the accurate mass and time tag approach, we identified and quantified ~1,100 islet proteins by at least two unique peptides with a false discovery rate
